Description
English: A reconstructed 3-D movie of a cell showing close association of viral particles with microtubules. BCBL-1 cells induced with TPA for 48 hours were fixed and permeablized. MTs and viral capsids were stained with mouse monoclonal anti-tubulin and rabbit-polyclonal anti-ORF65 antibodies, respectively, with anti-mouse IgG-Alexa Flour 488 (green) and anti-rabbit/IgG-Texas Red (red) secondary antibodies. Double staining was then examined under a confocal microscope (Leica TCS SPII confocal laser scanning system). A series of optical sections of the image were collected at increasing intervals of 0.32 µm from the bottom to the top. This was employed for the reconstruction of a 3-D representation of the specimen on the graphics computer with the LCS 3D software (Leica Microsystems).
