English: Representative interactions of 5C.C7 T cells retrovirally transduced to express the indicated sensors with CH27 B cell lymphoma APCs in the presence of MCC agonist peptide (10 M) are shown in S1 to S3 Videos. DIC images are shown on the top, with matching top-down, maximum projections of 3D sensor fluorescence data on the bottom. The sensor fluorescence intensity is displayed in a rainbow-like, false-color scale (increasing from blue to red). 20 s intervals in video acquisition are played back as 2 frames per second. The 5C.C7 T cell in S1 Video is transduced with SLP-76-GFP. Cell coupling occurs in frame 5 (2s indicated video time). A rapid transition from central to lamellal accumulation that subsequently fades more slowly is shown. |
