Ainda que o culto à Virgem de Guadalupe se tenha divulgado no território português logo após a batalha do Salado, em 1340, alguns elementos decorativos da Ermida, como os encordoados e figuras humanas dos capitéis, apresentam características tardo-góticas, o que indicia que o edifício remonta aos meados do século XV. Situada entre a Europa cristã e o Magrebe islâmico, o culto ganha importância devido aos frequentes confrontos com os mouros e à necessidade de resgate dos cristãos feitos cativos, já que o rei instituíra no Algarve o cargo de alfaqueque, instalando-o na zona para melhor poder negociar contrapartidas.
