Description
English: Two types of fibers from the surface of a Russian olive (Elaeagnus angustifolia) under microscope

Native, aqueous or temporary slides, as they are also called, are slides that last for a certain period of time and ordinary water is used for their preparation. They are easy and fast for preparation and do not require additional reagents, as well as are quite practical for observing certain structures under a microscope. The procedure for their preparation begins with the preparation of the object and cover glass, which should be wiped thoroughly. Two drops of water are placed on the object glass, one central large and one peripheral smaller. The wild olive leaf is taken. It can be noticed that the underside has a lighter green color. From the underside of the leaf, a razor blade scrapes off the silver covering of fibers onto the central drop of water. A cover glass is placed on it, which is lowered next to the peripheral drop of water and at an angle of 45° it moves towards the middle of the object glass until the moment when the two drops of water merge, during which it is lowered. This procedure needs to be performed precisely because otherwise air bubbles will form which will count during microscopy. In this way the native microscopic slide is prepared and it is ready for microscopy.

Врз предметното стакленце се ставаат две капки вода, една средишна поголема и една странична помала капка вода. Се зема листот од дива маслинка. Може да се забележи дека долната страна има посветло зелена боја, која потекнува од сребрената прекривка создадена од плочести и ѕвездовидни многуклеточни влакна. Од долната страна на листот со жилет се наросува од сребрената прекривка врз средишната капка со вода. Се става покровно стакленце кое се спушта покрај страничната капка со вода и под агол од 45° се движи кон средината на предметното стакленце се до моментот кога ќе се спојат двете капки со вода, при што се спушта. Оваа постапка потребно е да се изведе прецизно затоа што во спротивно ќе се образуваат воздушни меурчина кои ќе сметаат при микроскопирањето. На овој начин е подготвен нативниот микроскопски препарат и истиот е спремен за микроскопирање. Во видното поле на микроскоп се гледа плочесто и ѕвездовидно влакно едно до друго. И двата вида на покривни влакна се многуклеточни. Ѕвездовидното влакно има облик на ѕвезда, од каде доаѓа и неговото име. Секој крак од влакното претставува една клетка. Кај плочестиот вид на влакна, клетките се позбиени со своите странични ѕидови.
