La iconografía de la santa elegida por Gil de Siloe para el retablo no es original suya sino que sigue un tipo utilizado entonces en los talleres de Malinas; reúne en una misma figura dos tradiciones iconográficas distintas: la Santa Ana "maestra", reflejada con frecuencia sobre todo en las miniaturas de los libros de rezos manuscritos, que enseña a su hija las primeras letras en un libro, y la Santa Ana Triple que sostiene a su hija y a su nieto. La presencia del libro abierto aludiría también, para CJ Ara, "a su condición de mujer sabia transmisora del verdadero conocimiento". Doña Mencía de Mendoza tendría en torno a setenta o setenta y cinco años cuando encargó a Gil de Siloe el que podemos considerar "su testamento espiritual".
