Copyright (C) 2007 あら金。著作権はGFDLに寄託。 説明 C末端を活性化されたペプチドは隣接ペプチドのカルボニル酸素の付加脱離反応によりオキサゾリン体を生成する。オキサゾリン体の4位炭素はja:互変異性の平衡が存在しオキサゾール体を形成するためにラセミ化しやすい。その結果ペプチド合成法においてC末端を活性化するペプチド合成戦略はエピ化の問題を内包することになる。 なお、生体内でのペプチド合成ではC末端に増端するが、酵素反応であるため酵素と結合している瞬間だけカルボニルが活性化するだけなのでエピ化の問題が発生しない。 |
