English: Schematic representation of the genome and full-length cDNA clones of PVS isolate H95. PVS-H95 genome organization is shown at the top. The start and stop positions of each ORF (rectangle) are indicated with nucleotide numbers. Nucleotide substitutions among the eight full-length cDNA clones of PVS-H95 (bottom) are indicated with colored arrowheads; numbers indicate the nucleotide positions. A full-length cDNA clone pPVS-H-FL-AB was constructed by connecting eight clones (colored rectangles). Arrow indicates T7 promoter. Seven additional clones were constructed either by the replacement of a fragment produced by site-directed mutagenesis (e.g., pPVS-H-FL-V, −D, −E, -F) or from a different clone (e.g., pPVS-H-FL-C, −G, -H). The cDNA clone pPVS-H-FL-H with an asterisk repesents the consensus sequence of PVS-H95 genome. Nucleotides and coding amino acids in parentheses with a gray background vary from the consensus sequence of PVS-H95 genome. |
