Description
Deutsch: Die Abbildung zeigt in drei Schritten (I.,II., III.) schematisch den Ablauf der Methode cRACE, welche von Maruyama et al. (1995; PMID 7479016) veröffentlicht wurde.

I. Ein genspezifischer Primer (b) bindet spezifisch an einem mRNA-Molekül (a). II. Der Primer wird durch die Synthese der Erststrang-cDNA (c) verlängert.

III. Die einzelsträngige cDNA wird nach Beseitigung der RNA mit sich selbst ligiert, sodass eine Ring-Molekül entsteht. Mithilfe verschiedener genspezifischer Primer können anschließend verschachtelte Polymerasekettenreaktionen (PCR) für die Amplifikation des gesuchten Genabschnitts durchgeführt werden. Es sind mögliche Positionen für die Primer der ersten PCR (d) und für die Primer der zweiten PCR (e) eingezeichnet.
