English: DNA shuffling works through an ordered process of: (1) fragmentation of the parent sequence, (2) denaturation of dsDNA fragments to ssDNA fragments, (3) homology directed recombination, (4/5) primer-less PCR, and (6) isolating or repeating. The figure illustrates a simplified single sequence DNA shuffling walk-through. A DNA shuffling methodology using a gene family would differ only in the amount and diversity of starting sequences prior to fragmentation. Because the newly synthesized strands of DNA have a higher-than-normal mutational frequency, they are marked in black. |
