(a) Expressão heteróloga das proteínas de Rhodovulum sulfidophilum sob condições foto-heterotróficas. um R. sulfidophilum recombinante abrigando o vetor de ampla gama de hospedeiros pBBR1MCS-2 com um domínio repetitivo MaSp1 de Nephila clavipes foi desenvolvido para expressar a proteína da seda da linha de aranha. (b) Um cassete de gene contendo o promotor trc (Ptrc) e MaSp1- (1-mer, 2-mer, 3-mer, e 6-mer) foi inserido em pBBR1MCS-2, e uma histidine tag estava presente no terminal N de MaSp1 (caixa de cor rosa). (c) sistema SDS-PAGE com tampão Tricina (16,5%) das proteínas solúveis de quatro dias de culturas recombinantes de R. sulfidophilum. (d) Western blot usando anticorpo monoclonal anti-His Tag, que tem como alvo proteínas MaSp1- (1-mer, 2-mer, 3-mer ou 6-mer) marcadas com histidina (FOONG et. al., 2020). |
