מצפה אשתמוע נמצא בהר חברון, מצפון ובצמוד ליישוב שמעה וממערב לכפר הערבי א-סמוע. המאחז שייך למועצה אזורית הר חברון. המאחז הוקם בשנת 2002 על ידי קבוצת בחורי ישיבה מהיישוב עתניאל, ושמו ניתן לו כיוון שהוא צופה אל המקום בו מעריכים כי התקיימה עיר הלווים אשתמוע.
בתחילה הוקם המאחז באמצע הדרך בין היישוב עתניאל לישוב שמעה, כדי ליצור רצף התיישבות יהודית מכיוון דרום לעתניאל. בעקבות הסדר עם משרד הביטחון, פונה המאחז בהסכמה במרחשוון התשס"ג בתמורה להכרה חוקית בנקודה הנעזבת, והוקם זמנית בנקודה הנוכחית בשטחו של היישוב שמעה. היעוד הראשוני של הקבוצה היה הקמת ישוב מעורב לדתיים וחילוניים ברוח הרב קוק.
בשיאו מנה הגרעין כ-10 משפחות, אך בעקבות ההתנגדות הפוליטית והקשיים של תקופת ההתנתקות, נגנז רעיון היישוב והגרעין התפרק
