Description
English: Supplemental Video 1. Localization and motility of fluorescent T and B cells in the popliteal LN of a SCID mouse following their transfer from arthritic BALB/c mice. Donor T cells were labeled with CellTracker Red (CMTPX) and non-T cells (~90% B cells) with CellTracker Green (CMFDA), and mixed at a ratio of 1:2 prior to injection into the SCID mouse. The popliteal LN was subjected to TPM imaging on day 7 after cell transfer. Z series of images were acquired from the LN in vivo. Autofluorescent subcapsular sinus macrophages (light blue) are seen in the top layer of the cortex. B cells (green) localize in the more superficial, while T cells (red) in the deeper areas of the LN. Most T and B cells exhibit a polarized shape and 'random' movement. Imaging depth: 87 μm; time elapsed: 9.5 min.
