English: Localized synthesis of PI 3,4,5-P3 at the sites of chlamydial entry. Cos-7 cells expressing the pleckstrin homology domain of Bruton's tyrosine kinase fused to green fluorescent protein (BTK-PH-GFP) were monitored by live cell microscopy upon infection with CMTPX-labeled C. trachomatis serovar L2 elementary bodies. Images were acquired at 5 s intervals and compiled using the NIH Image J software. Note that the recruitment of the fluorescent probe is short-lived, indicating the transient nature of the signaling cascade. It is important to note that not all of the EB particles induced the synthesis of PI 3,4,5-P3 because there likely were non-infectious particles in the EB preparations. Loss of infectivity occurs during harvest, purification, and labeling of elementary bodies. A) Mock-treated cells; B) Wortmannin (100 nM)-treated cells. |
