As origens da Igreja Matriz de Vila do Bispo remontam ao século XVI. No entanto, o edifício atual resultou de uma profunda remodelação levada a cabo nos finais do século XVII e no início do século XVIII. O corpo da igreja possui uma só nave, sendo a capela-mor antecedida por um arco triunfal. A ornamentação interior deste templo data maioritariamente do primeiro quartel do século XVIII, pelo que a igreja apresenta uma quase perfeita unidade estética, predominando os elementos decorativos de estilo barroco. Entre os retábulos deste templo, o destaque vai para o exemplar que se encontra na capela-mor e o retábulo da Capela de Nossa Senhora do Carmo, ambos do período joanino. Entre os tesouros artísticos desta igreja, merecem particular referência a imagem quinhentista de Nossa Senhora da Conceição, uma naveta do século XV e ainda duas tábuas pintadas, representando São Pedro e São Paulo. Uma das características mais interessantes desta igreja é a cobertura da nave, que utiliza pintura de brutescos em caixotões.
