English: SH-SY5Y/Neo cells were loaded with TMRM as indicated above and exposed to ionomycin (2 µM). Images were acquired using a fluorescence inverted microscope (Nikon Eclipse TE-2000-S, Birlingam, CA, USA) equipped with a 150 W Xenon lamp and 100×, 1.3 numerical aperture, epifluorescence oil immersion objective and commercial software (Metamorph, Universal Imaging Corporation, Silicon Valley, CA, USA). Frames were acquired every 15 seconds over 10 minutes. Fluorescence was observed using an excitation filter of 535 nm and an emission filter of 590 nm. |
